I also think the language of the Nature article re genetic therapies in the pipeline is interesting to read ... Maybe these pipeline lists will swell in upcoming months ... glad the FDA seems to be 'on it' ... Just read Nobel Laureate Ishiguro's "Klara and the Sun" about Artificial Intelligence Friends, Artificial Friends, Robot People, and Personal Robots, where the main human character was also 'lifted' - genetically engineered for enhancement because her parents had the money (in Britain?) ... but which makes her feel sick much of the time (so fallen in a sense too) ... And, no, it doesn't really interest me that much. I see it as a mirage. You know yourself, as a yoga teacher, that as human beings our reality lies in the Spiritual World, from which we are having a temporary physical experience. Why we would wish to prolong that experience baffles me. I think the answer is pure egotism: I wanna be ME in this physical body as long as I can.

Such an approach is not for me!

Happy Birthday and 2022! And, no, it doesn't really interest me that much. I see it as a mirage. You know yourself, as a yoga teacher, that as human beings our reality lies in the Spiritual World, from which we are having a temporary physical experience. Why we would wish to prolong that experience baffles me. I think the answer is pure egotism: I wanna be ME in this physical body as long as I can. Such an approach is not for me! How are you? How did your regaling of Princeton students about Yemen and the Middle East go (and regarding the new related university at the US State Department even :)? What are you doing over Xmas and New Year? Might you be around even, or heading to Yemen and the MIddle East, or warmer climes south, or ... ?

... hoping you're navigating the oncoming winter in the DC-area alright. It looks like I'll be heading back to Pgh for Xmas, :) but only for a brief trip, so may not head to DC as I've done in some previous years, although it would be great to see you somehow, either possibly even to celebrate Xmas together in a video call, ... or even if my plans change, and I head back to Pgh, and with a trip to DC in between Xmas, and then heading back to the SF Bay Area :) It was good to talk with my mother last night (see below), having invited her out here for Xmas, but she may not be doing this for awhile, since it's a different trip to California than to Portland, Maine, for Thanksgiving (where she went to visit Sandy) culturally perhaps somehow too. 

Am seeking 1 or 2 new Scottish Small Pipes' students for online learning currently :) Know anyone who might be interested?

Just searched on and found a bit surprisingly these 2 somewhat new articles on aging reversal (below), where the focus is on research to "rejuvenate human cells by reprogramming them back to a youthful state" ... I'm wondering if there are a zillion new scientific opportunities in some of these aging reversal genetic drug therapies emerging regards, thanks to the past 18 months of dramatic, I hypothesize, vaccine developments and related fundamental research (at the NIH in scaling organizationally too). The NYT's Wade article is a good overview, and with an angle on rejuvenation of cells, and its related aging reversal history, and key scientific ideas ...  

Am wondering too about Actual-Virtual, Physical-Digital experiments' history, in broad strokes, and even regarding a scientific literature review, and possible comparative papers - in many disciplines of 'hard science' from biology to physics to chemistry to geology to materials science to psychology (re rigorous experimental paradigm) ... and what we could learn from this for a realistic virtual earth for STEM for Genetics ... and especially for aging reversal genetic drug therapies ... (e.g. in Google Street View with time slider at the cellular and molecular levels, how do particles and molecules rearrange themselves as aging reversal hypothetically takes place ... and with ever expanding data sets, and machine learning even?). But thinking about classic scientific experiments, whether they be in animal behavior, re BF Skinner's studies with pigeons and rats, or atmospheric gases' experiments, ...  or a vast myriad of other examples of scientific experiments, from some decades ago even, how have they emerged anew with computer simulations, modeling and similar?

Now how might you or I begin to play with aging reversal basic science and in a realistic virtual earth for STEM ... at the molecular or cellular levels, and in a Do-It-Yourself DIY way? And to even edit or transform our own genome, perhaps aging reversal wise ... and via even WikiTree genealogy software, I wonder, as an entry way into this self-genetic-engineering ... and with realistic virtual avatar bots too? :) ... https://www.wikitree.com/wiki/MacLeod-2524 ... say working with George Church collaboratively on these questions too? 

One 'definition' of intelligence, I recall from family friend George Alexander MD is 'problem solving' (and scientific experiments do this), and another, though, is modeling ... I am seeking for a realistic virtual earth to perhaps facilitate this latter approach to intelligence in myriad excellent iterating ways, and digitally or virtually and with computing and machine learning even :)

Researchers at Stanford University report that they can rejuvenate human cells by reprogramming them back to a youthful state. They hope that the technique will help in the treatment of diseases, such as osteoarthritis and muscle wasting, that are caused by the aging of tissue cells.

A major cause of aging is thought to be the errors that accumulate in the epigenome, the system of proteins that packages the DNA and controls access to its genes. The Stanford team, led by Tapash Jay Sarkar, Dr. Thomas A. Rando and Vittorio Sebastiano, say their method, designed to reverse these errors and walk back the cells to their youthful state, does indeed restore the cells’ vigor and eliminate signs of aging.

In their report, published on Tuesday in Nature Communications, they described their technique as “a significant step toward the goal of reversing cellular aging” and could produce therapies “for aging and aging-related diseases.”

Leonard P. Guarente, an expert on aging at M.I.T., said the method was “one of the most promising areas of aging research” but that it would take a long time to develop drugs based on RNA, the required chemical.

The Stanford approach utilizes powerful agents known as Yamanaka factors, which reprogram a cell’s epigenome to its time zero, or embryonic state.

Embryonic cells, derived from the fertilized egg, can develop into any of the specialized cell types of the body. Their fate, whether to become a skin or eye or liver cell, is determined by chemical groups, or marks, that are tagged on to their epigenome.

In each type of cell, these marks make accessible only the genes that the cell type needs, while locking down all other genes in the DNAs. The pattern of marks thus establishes each cell’s identity.

As the cell ages, it accumulates errors in the marking system, which degrade the cell’s efficiency at switching on and off the genes needed for its operations.

In 2006 Dr. Shinya Yamanaka, a stem-cell researcher at Kyoto University, amazed biologists by showing that a cell’s fate could be reversed with a set of four transcription factors — agents that activate genes — that he had identified. A cell dosed with the Yamanaka factors erases the marks on the epigenome, so the cell loses its identity and reverts to the embryonic state. Erroneous marks gathered during aging are also lost in the process, restoring the cell to its state of youth. Dr. Yamanaka shared the 2012 Nobel Prize in medicine for the work.


In 2006, Dr. Shinya Yamanaka, a stem cell researcher and Nobel laureate, identified four transcription factors that can be manipulated to reverse aging in cells.

But the Yamanaka factors are no simple panacea. Applied to whole mice, the factors made cells lose their functions and primed them for rapid growth, usually cancerous; the mice all died.

In 2016, Juan Carlos Izpisua Belmonte, of the Salk Institute for Biological Studies in San Diego, found that the two effects of the Yamanaka factors — erasing cell identity and reversing aging — could be separated, with a lower dose securing just age reversal. But he achieved this by genetically engineering mice, a technique not usable in people.

In their paper on Tuesday, the Stanford team described a feasible way to deliver Yamanaka factors to cells taken from patients, by dosing cells kept in cultures with small amounts of the factors.

If dosed for a short enough time, the team reported, the cells retained their identity but returned to a youthful state, as judged by several measures of cell vigor.

Dr. Sebastian said the Yamanaka factors appeared to operate in two stages, as if they were raising the epigenome’s energy to one level, at which the marks of aging were lost, and then to a higher level at which cell identity was erased.

The Stanford team extracted aged cartilage cells from patients with osteoarthritis and found that after a low dosage of Yamanaka factors the cells no longer secreted the inflammatory factors that provoke the disease. The team also found that human muscle stem cells, which are impaired in a muscle-wasting disease, could be restored to youth. Members of the Stanford team have formed a company, Turn Biotechnologies, to develop therapies for osteoarthritis and other diseases.

The study is “definitively a step forward in the goal of reversing cellular aging,” Dr. Izpisua Belmonte said.
